To effectively guide agricultural management planning strategies and policy, it is important to simulate water quantity and quality patterns and quantify the impact of land use and climate change on underlying processes. Environmental models that depict alterations in surface and groundwater quality and quantity at a catchment scale require substantial input, particularly concerning movement and retention in the unsaturated zone. Over the past few decades, numerous soil information sources, containing structured data on diverse basic and advanced soil parameters, alongside innovative solutions to estimate missing soil data, have become increasingly available. This study aims to: i) catalogue open-source soil datasets and pedotransfer functions (PTFs) applicable in simulation studies across European catchments, ii) evaluate the performance of selected PTFs and iii) present compiled R scripts proposing estimation solutions to address soil physical, hydraulic, and chemical soil data needs and gaps in catchment-scale environmental modelling in Europe. Our focus encompassed basic soil properties, bulk density, porosity, albedo, soil erodibility factor, field capacity, wilting point, available water capacity, saturated hydraulic conductivity, and phosphorus content. We aim to recommend widely supported data sources and pioneering prediction methods that maintain physical consistency, and present them through streamlined workflows.

Targeted drug delivery is a promising approach for many serious diseases, such as glioblastoma multiforme, one of the most common and devastating brain tumor. In this context, this work addresses the optimization of the controlled release of drugs which are carried by extracellular vesicles. Towards this goal, we derive and numerically verify an analytical solution for the end-to-end system model. We then apply the analytical solution either to reduce the disease treatment time or to reduce the amount of required drugs. The latter is formulated as a bilevel optimization problem, whose quasiconvex/quasiconcave property is proved here. For solving the optimization problem, we propose and utilize a combination of bisection method and golden-section search. The numerical results demonstrate that the optimization can significantly reduce the treatment time and/or the required drugs carried by extracellular vesicles for a therapy compared to the steady state solution.

Globally, significant societal resources are devoted to mitigating negative effects of eutrophication from excessive phosphorus (P) and nitrogen (N) loading. Potential effectiveness of mitigation measures and possible confounding factors are often assessed using studies conducted in headwater catchments. However, success is often evaluated based on trends in river mouth water chemistry. It is not clear how transferrable insights from headwater catchments are to larger rivers. Here, relationships between P and suspended solids (SS) identified in small agricultural headwater catchments were applied to 30 larger, mixed land use catchments draining into Mälaren, a Swedish great lake. The approach is based on Oblique Predictive Clustering Trees (SPYCTs). Interpretation of the model predictions through the SHAP method. Validation of the model performed on more than 1000 samples of job seekers data. The model supports job seekers in finding optimal career solutions. Abstract We address the problem of estimating the time-to-employment of a jobseeker using survival analysis and oblique predictive clustering tree. Unlike standard survival analysis, oblique predictive clustering tree can handle categorical and continuous data and is capable of modelling non-linear dependences. Treating the censored data as missing data opens the possibility to perform survival analysis by using structured output prediction in semi-supervised multi-target regression setting. The effectiveness of this approach is shown on a real dataset from Public Employment Services in Slovenia, comprising time-to-employment records with jobseekers’ personal and professional characteristics. The performances are compared with six state-of-the-art AI methods. To the best of our knowledge, this is the first example of using semi-supervised oblique predictive clustering tree for survival analysis.

The goal was to gather data for defining the user needs for a discovery platform for Social Sciences and Humanities and for building Personas and Scenarios. The data was collected in the period 01/10/2019 - 30/03/2020. This is a first version of the data. A more complete version will be uploaded by the end of the project (March 2023), and it will include additional data, at which point this version will be considered obsolete.

Its latest version (SWAT+) adds new capabilities to the tool, and collectively with increasing computational power and availability of public datasets expands model complexity as well as provides pathways for serious errors in the model setup process. These models are used to run scenarios to support decision-making processes, hence undetected faults can have a substantial socio-economic and environmental impact. We propose a 5-step SWAT+ model setup verification workflow assessing the soundness of processes related to weather, water balance, management, plant growth, point source and tile drain flows. We developed an R package, called SWATdoctR, which guides the user through the model setup verification process, allowing the identification of typical, but repeatedly overlooked, issues. The workflow and the functionality of the tool is demonstrated in 4 SWAT+ setups in different catchments, at various stages of model setup.

In recent years, many pre-clinical studies have tested gene therapy approaches as possible treatments for epilepsy, following the idea that they may provide an alternative to conventional pharmacological and surgical options. Multiple gene therapy approaches have been developed, including those based on anti-sense oligonucleotides, RNA interference, and viral vectors. In this opinion article, we focus on translational issues related to viral vector-mediated gene therapy for epilepsy. Research has advanced dramatically in addressing issues like viral vector optimization, target identification, strategies of gene expression, editing or regulation, and safety. Some of these pre-clinically validated potential gene therapies are now being tested in clinical trials, in patients with genetic or focal forms of drug-resistant epilepsy. Here, we discuss the ongoing translational research and the advancements that are needed and expected in the near future. We then describe the clinical trials in the pipeline and the further challenges that will need to be addressed at the clinical and economic levels. Our optimistic view is that all these issues and challenges can be overcome, and that gene therapy approaches for epilepsy will soon become a clinical reality.

To effectively guide agricultural management planning strategies and policy, it is important to simulate water quantity and quality patterns and quantify the impact of land use and climate change on underlying processes. Environmental models that depict alterations in surface and groundwater quality and quantity at a catchment scale require substantial input, particularly concerning movement and retention in the unsaturated zone. Over the past few decades, numerous soil information sources, containing structured data on diverse basic and advanced soil parameters, alongside innovative solutions to estimate missing soil data, have become increasingly available. This study aims to: i) catalogue open-source soil datasets and pedotransfer functions (PTFs) applicable in simulation studies across European catchments, ii) evaluate the performance of selected PTFs and iii) present compiled R scripts proposing estimation solutions to address soil physical, hydraulic, and chemical soil data needs and gaps in catchment-scale environmental modelling in Europe. Our focus encompassed basic soil properties, bulk density, porosity, albedo, soil erodibility factor, field capacity, wilting point, available water capacity, saturated hydraulic conductivity, and phosphorus content. We aim to recommend widely supported data sources and pioneering prediction methods that maintain physical consistency, and present them through streamlined workflows.

Targeted drug delivery is a promising approach for many serious diseases, such as glioblastoma multiforme, one of the most common and devastating brain tumor. In this context, this work addresses the optimization of the controlled release of drugs which are carried by extracellular vesicles. Towards this goal, we derive and numerically verify an analytical solution for the end-to-end system model. We then apply the analytical solution either to reduce the disease treatment time or to reduce the amount of required drugs. The latter is formulated as a bilevel optimization problem, whose quasiconvex/quasiconcave property is proved here. For solving the optimization problem, we propose and utilize a combination of bisection method and golden-section search. The numerical results demonstrate that the optimization can significantly reduce the treatment time and/or the required drugs carried by extracellular vesicles for a therapy compared to the steady state solution.

Globally, significant societal resources are devoted to mitigating negative effects of eutrophication from excessive phosphorus (P) and nitrogen (N) loading. Potential effectiveness of mitigation measures and possible confounding factors are often assessed using studies conducted in headwater catchments. However, success is often evaluated based on trends in river mouth water chemistry. It is not clear how transferrable insights from headwater catchments are to larger rivers. Here, relationships between P and suspended solids (SS) identified in small agricultural headwater catchments were applied to 30 larger, mixed land use catchments draining into Mälaren, a Swedish great lake. The approach is based on Oblique Predictive Clustering Trees (SPYCTs). Interpretation of the model predictions through the SHAP method. Validation of the model performed on more than 1000 samples of job seekers data. The model supports job seekers in finding optimal career solutions. Abstract We address the problem of estimating the time-to-employment of a jobseeker using survival analysis and oblique predictive clustering tree. Unlike standard survival analysis, oblique predictive clustering tree can handle categorical and continuous data and is capable of modelling non-linear dependences. Treating the censored data as missing data opens the possibility to perform survival analysis by using structured output prediction in semi-supervised multi-target regression setting. The effectiveness of this approach is shown on a real dataset from Public Employment Services in Slovenia, comprising time-to-employment records with jobseekers’ personal and professional characteristics. The performances are compared with six state-of-the-art AI methods. To the best of our knowledge, this is the first example of using semi-supervised oblique predictive clustering tree for survival analysis.

A more complete version will be uploaded by the end of the project (March 2023), and it will include additional data, at which point this version will be considered obsolete.

Its latest version (SWAT+) adds new capabilities to the tool, and collectively with increasing computational power and availability of public datasets expands model complexity as well as provides pathways for serious errors in the model setup process. These models are used to run scenarios to support decision-making processes, hence undetected faults can have a substantial socio-economic and environmental impact. We propose a 5-step SWAT+ model setup verification workflow assessing the soundness of processes related to weather, water balance, management, plant growth, point source and tile drain flows. We developed an R package, called SWATdoctR, which guides the user through the model setup verification process, allowing the identification of typical, but repeatedly overlooked, issues. The workflow and the functionality of the tool is demonstrated in 4 SWAT+ setups in different catchments, at various stages of model setup.

In recent years, many pre-clinical studies have tested gene therapy approaches as possible treatments for epilepsy, following the idea that they may provide an alternative to conventional pharmacological and surgical options. Multiple gene therapy approaches have been developed, including those based on anti-sense oligonucleotides, RNA interference, and viral vectors. In this opinion article, we focus on translational issues related to viral vector-mediated gene therapy for epilepsy. Research has advanced dramatically in addressing issues like viral vector optimization, target identification, strategies of gene expression, editing or regulation, and safety. Some of these pre-clinically validated potential gene therapies are now being tested in clinical trials, in patients with genetic or focal forms of drug-resistant epilepsy. Here, we discuss the ongoing translational research and the advancements that are needed and expected in the near future. We then describe the clinical trials in the pipeline and the further challenges that will need to be addressed at the clinical and economic levels. Our optimistic view is that all these issues and challenges can be overcome, and that gene therapy approaches for epilepsy will soon become a clinical reality.
