International audience; Highly unsaturated fatty acids of the omega-3 series (HUFA) are major constituents of cell membranes. yet are poorly synthesised de novo by consumers. Their production, mainly supported by aquatic microalgae, has been decreasing with global change. The consequences of such reductions may be profound for ectotherm consumers, as temperature tightly regulates the HUFA content in cell membranes, maintaining their functionality. Integrating individual, tissue and molecular approaches, we examined the consequences of the combined effects of temperature and HUFA depletion on the key cardio-respiratory functions of the golden grey mullet, an ectotherm grazer of high ecological importance. For 4 months, fish were exposed to two contrasting HUFA diets [4.8% eicosapentaenoic acid (EPA)+docosahexaenoic acid (DHA) on dry matter (DM) versus 0.2% EPA+DHA on DM] at 12 and 20 degrees C. Ventricular force development coupled with gene expression profiles measured on cardiac muscle suggest that combining HUFA depletion with warmer temperatures leads to: (1) a proliferation of sarcolemmal and sarcoplasmic reticulum Ca2+ channels and (2) a higher force-generating ability by increasing extracellular Ca2+ influx via sarcolemmal channels when the heart has to sustain excessive effort due to stress and/or exercise. At the individual scale, these responses were associated with a greater aerobic scope, maximum metabolic rate and net cost of locomotion, suggesting the higher energy cost of this strategy. This impaired cardiac performance could have wider consequences for other physiological performance such as growth, reproduction or migration, all of which greatly depend on heart function.

AbstractDuring early stages of development vertebrates rely on an immature immune system to fight pathogens, but in non mammalian species few studies have taken an in-depth analysis of the transition from reliance on innate immune mechanisms to the appearance of adaptive immunity. Using rainbow trout as a model we characterized responses to two natural pathogens of this species, the Gram negative bacterium Aeromonas salmonicida and the virus VHSV, using microarray analysis at four early life history stages; eyed egg, post hatch, first feeding and three weeks post first feeding when adaptive immunity starts to be effective. All stages responded to both infections, but the complexity of the response increased with developmental stage. The response to virus showed a clear interferon response only from first feeding. In contrast, bacterial infection induced a marked response from early stages, with modulation of inflammatory, antimicrobial peptide and complement genes across all developmental stages. Whilst the viral and bacterial responses were distinct, there were modulated genes in common, mainly of general inflammatory molecules. This work provides a first platform to explore the development of fish immunity to infection and to compare the age-dependent changes (from embryo to adults) across vertebrates.

Abstract Chicken genome mapping is important for a range of scientific disciplines. The ability to distinguish chromosomes of the chicken and other birds is thus a priority. Here we describe the molecular cytogenetic characterization of each chicken chromosome using chromosome painting and mapping of individual clones by FISH. Where possible, we have assigned the chromosomes to known linkage groups. We propose, on the basis of size, that the NOR chromosome is approximately the size of chromosome 22; however, we suggest that its original assignment of 16 should be retained. We also suggest a definitive chromosome classification system and propose that the probes developed here will find wide utility in the fields of developmental biology, DT40 studies, agriculture, vertebrate genome organization, and comparative mapping of avian species.

Roland; François De Vleeschouwer;International audience; Remote Northern Hemisphere (NH) and Southern Hemisphere (SH) lake sediment and peat records of mercury (Hg) deposition have shown a ×3 to ×5 Hg enrichment since pre-industrial times (<1880 AD), leading to the perception that global atmospheric Hg enrichment is moderate and uniform across the hemispheres. Anthropogenic Hg emissions in the NH are, however, approximately 4 times higher than those in the SH. Here, we reconstruct atmospheric Hg deposition in four remote SH peatlands and review sediment and peat Hg records from both hemispheres. We observe a ×4 all-time enrichment in SH Hg deposition from pre-anthropogenic (<1450 AD) to the late 20th-century periods, which is lower than the large ×16 all-time enrichment in NH Hg deposition. We attribute this difference to lower anthropogenic Hg emissions in the SH, and higher natural atmospheric SH Hg concentrations, supported by ×2 higher natural background Hg accumulation in SH peat records. We suggest that the higher SH natural atmospheric Hg deposition reflects the SH land–ocean distribution, and is driven by important SH marine Hg emissions. Our findings indicate that atmospheric Hg background levels and anthropogenic enrichment in both hemispheres are different and should be taken into account in international Hg assessments and environmental policy.

Detected volatile constituents in studied species were constituted mainly by sesquiterpene, as well as diterpene compounds. All so-established compositions cannot successfully index some of them to known chemotypes but afforded the discovery of new chemotypes such as cuparane/fusicoccane. The major component of B. francana was isolated and characterized as a new zierane-type sesquiterpene called ziera-12(13),10(14)-dien-5-ol (23). In addition, qualitative intraspecies variations of chemical composition were very important particularly for B. francana which possessed three clearly defined different compositions. We report here also the first phytochemical investigation of Acromastigum species. Moreover, crude diethyl ether extract of B. vitatta afforded a new bis(bibenzyl) called vittatin (51), for which a putative biosynthesis was suggested.

E.; Metzl, Nicolas; Olsen, Are; Sabine, Christopher L.; Heimann, M.;International audience; A temporally and spatially resolved estimate of the global surface-ocean CO2 partial pressure field and the sea-air CO2 flux is presented, obtained by fitting a simple data-driven diagnostic model of ocean mixed-layer biogeochemistry to surface-ocean CO2 partial pressure data from the SOCAT v1.5 database. Results include seasonal, interannual, and short-term (daily) variations. In most regions, estimated seasonality is well constrained from the data, and compares well to the widely used monthly climatology by Takahashi et al. (2009). Comparison to independent data tentatively supports the slightly higher seasonal variations in our estimates in some areas. We also fitted the diagnostic model to atmospheric CO2 data. The results of this are less robust, but in those areas where atmospheric signals are not strongly influenced by land flux variability, their seasonality is nevertheless consistent with the results based on surface-ocean data. From a comparison with an independent seasonal climatology of surface-ocean nutrient concentration, the diagnostic model is shown to capture relevant surface-ocean biogeochemical processes reasonably well. Estimated interannual variations will be presented and discussed in a companion paper.

Knowledge about HNS monitoring using radar remote sensing is not as extensive as for oil spills; however, any progress on this issue would likely advance the monitoring of both chemical and oil-related incidents. To address the need for HNS monitoring, an experiment was conducted in May 2015 over the Mediterranean Sea during which controlled releases of HNS were imaged by a multifrequency radar system. The aim of this experiment was to establish a procedure for collecting evidence of illegal maritime pollution by noxious liquid substances using airborne radar sensors. In this paper, we demonstrate the ability of radar imagery to detect and characterize chemicals at sea. A normalized polarization difference parameter is introduced to quantify both the impacts of released product on the ocean surface and the relative concentration of the substance within the spill. We show that radar imagery can provide knowledge of the involved HNS. In particular, one can distinguish a product that forms a film on the top of the sea surface from another that mixes with seawater, the information that is critical for efficient cleanup operations.

Our first objective was to test the effect of dietary carbohydrates themselves (without modification of dietary protein intake) on hepatic glucose gene expression (taking into account the paralogs). The second aim was to research if two isogenic trout lines had different responses to carbohydrate intake, showing one with a better use dietary carbohydrates. Thus, we used two isogenic lines of rainbow trout (named A32h and AB1h) fed with either a high carbohydrate diet or a low carbohydrate diet for 12 weeks. We analysed the zootechnical parameters, the plasma metabolites, the hepatic glucose metabolism at the molecular level and the hormonal-nutrient sensing pathway. Globally, dietary carbohydrate intake was associated with hyperglycaemia and down regulation of the energy sensor Ampk, but also with atypical regulation of glycolysis and gluconeogenesis in the liver. Indeed, the first steps of glycolysis and gluconeogenesis catalysed by the glucokinase and the phospenolpyruvate carboxykinase are regulated at the molecular level by dietary carbohydrates as expected (i.e. induction of the glycolytic gck and repression of the gluconeogenic pck); by contrast, and surprisingly, for two other key glycolytic enzymes (phosphofructokinase enzyme – pfkl and pyruvate kinase – pk) some of the paralogs (pfklb and pklr) are inhibited by carbohydrates whereas some of the genes coding gluconeogenic enzymes (the glucose-6-phosphatase enzyme g6pcb1b and g6pcb2a gene and the fructose1-6 biphosphatase paralog fbp1a) are induced. On the other hand, some differences for the zootechnical parameters and metabolic genes were also found between the two isogenic lines, confirming the existence of genetic polymorphisms for nutritional regulation of intermediary metabolism in rainbow trout. In conclusion, our study determines some new and unexpected molecular regulations of the glucose metabolism in rainbow trout which may partly lead to the poor utilization of dietary carbohydrates and it underlines the existence of differences in molecular regulation of glucose metabolism between two isogenic lines which provides arguments for future selection of rainbow trout. Summary: Using isogenic lines, this study determines some new, unexpected molecular regulation of the glucose metabolism in rainbow trout, which may partly lead to the poor utilization of dietary carbohydrates.

Models predict that some portion of this release will be offset by increased production of Arctic and boreal biomass; however, the lack of robust estimates of net carbon balance increases the risk of further overshooting international emissions targets. Precise empirical or model-based assessments of the critical factors driving carbon balance are unlikely in the near future, so to address this gap, we present estimates from 98 permafrost-region experts of the response of biomass, wild␣re, and hydrologic carbon ␣ux to climate change. Results suggest that contrary to model projections, total permafrost-region biomass could decrease due to water stress and disturbance, factors that are not adequately incorporated in current models. Assessments indicate that end-of-the-century organic carbon release from Arctic rivers and collapsing coastlines could increase by 75% while carbon loss via burning could increase four-fold. Experts identi␣ed water balance, shifts in vegetation community, and permafrost degradation as the key sources of uncertainty in predicting future system response. In combination with previous ␣ndings, results suggest the permafrost region will become a carbon source to the atmosphere by 2100 regardless of warming scenario but that 65%–85% of permafrost carbon release can still be avoided if human emissions are actively reduced.

International audience; Highly unsaturated fatty acids of the omega-3 series (HUFA) are major constituents of cell membranes. yet are poorly synthesised de novo by consumers. Their production, mainly supported by aquatic microalgae, has been decreasing with global change. The consequences of such reductions may be profound for ectotherm consumers, as temperature tightly regulates the HUFA content in cell membranes, maintaining their functionality. Integrating individual, tissue and molecular approaches, we examined the consequences of the combined effects of temperature and HUFA depletion on the key cardio-respiratory functions of the golden grey mullet, an ectotherm grazer of high ecological importance. For 4 months, fish were exposed to two contrasting HUFA diets [4.8% eicosapentaenoic acid (EPA)+docosahexaenoic acid (DHA) on dry matter (DM) versus 0.2% EPA+DHA on DM] at 12 and 20 degrees C. Ventricular force development coupled with gene expression profiles measured on cardiac muscle suggest that combining HUFA depletion with warmer temperatures leads to: (1) a proliferation of sarcolemmal and sarcoplasmic reticulum Ca2+ channels and (2) a higher force-generating ability by increasing extracellular Ca2+ influx via sarcolemmal channels when the heart has to sustain excessive effort due to stress and/or exercise. At the individual scale, these responses were associated with a greater aerobic scope, maximum metabolic rate and net cost of locomotion, suggesting the higher energy cost of this strategy. This impaired cardiac performance could have wider consequences for other physiological performance such as growth, reproduction or migration, all of which greatly depend on heart function.

AbstractDuring early stages of development vertebrates rely on an immature immune system to fight pathogens, but in non mammalian species few studies have taken an in-depth analysis of the transition from reliance on innate immune mechanisms to the appearance of adaptive immunity. Using rainbow trout as a model we characterized responses to two natural pathogens of this species, the Gram negative bacterium Aeromonas salmonicida and the virus VHSV, using microarray analysis at four early life history stages; eyed egg, post hatch, first feeding and three weeks post first feeding when adaptive immunity starts to be effective. All stages responded to both infections, but the complexity of the response increased with developmental stage. The response to virus showed a clear interferon response only from first feeding. In contrast, bacterial infection induced a marked response from early stages, with modulation of inflammatory, antimicrobial peptide and complement genes across all developmental stages. Whilst the viral and bacterial responses were distinct, there were modulated genes in common, mainly of general inflammatory molecules. This work provides a first platform to explore the development of fish immunity to infection and to compare the age-dependent changes (from embryo to adults) across vertebrates.

Abstract Chicken genome mapping is important for a range of scientific disciplines. The ability to distinguish chromosomes of the chicken and other birds is thus a priority. Here we describe the molecular cytogenetic characterization of each chicken chromosome using chromosome painting and mapping of individual clones by FISH. Where possible, we have assigned the chromosomes to known linkage groups. We propose, on the basis of size, that the NOR chromosome is approximately the size of chromosome 22; however, we suggest that its original assignment of 16 should be retained. We also suggest a definitive chromosome classification system and propose that the probes developed here will find wide utility in the fields of developmental biology, DT40 studies, agriculture, vertebrate genome organization, and comparative mapping of avian species.

Roland; François De Vleeschouwer;International audience; Remote Northern Hemisphere (NH) and Southern Hemisphere (SH) lake sediment and peat records of mercury (Hg) deposition have shown a ×3 to ×5 Hg enrichment since pre-industrial times (<1880 AD), leading to the perception that global atmospheric Hg enrichment is moderate and uniform across the hemispheres. Anthropogenic Hg emissions in the NH are, however, approximately 4 times higher than those in the SH. Here, we reconstruct atmospheric Hg deposition in four remote SH peatlands and review sediment and peat Hg records from both hemispheres. We observe a ×4 all-time enrichment in SH Hg deposition from pre-anthropogenic (<1450 AD) to the late 20th-century periods, which is lower than the large ×16 all-time enrichment in NH Hg deposition. We attribute this difference to lower anthropogenic Hg emissions in the SH, and higher natural atmospheric SH Hg concentrations, supported by ×2 higher natural background Hg accumulation in SH peat records. We suggest that the higher SH natural atmospheric Hg deposition reflects the SH land–ocean distribution, and is driven by important SH marine Hg emissions. Our findings indicate that atmospheric Hg background levels and anthropogenic enrichment in both hemispheres are different and should be taken into account in international Hg assessments and environmental policy.

Detected volatile constituents in studied species were constituted mainly by sesquiterpene, as well as diterpene compounds. All so-established compositions cannot successfully index some of them to known chemotypes but afforded the discovery of new chemotypes such as cuparane/fusicoccane. The major component of B. francana was isolated and characterized as a new zierane-type sesquiterpene called ziera-12(13),10(14)-dien-5-ol (23). In addition, qualitative intraspecies variations of chemical composition were very important particularly for B. francana which possessed three clearly defined different compositions. We report here also the first phytochemical investigation of Acromastigum species. Moreover, crude diethyl ether extract of B. vitatta afforded a new bis(bibenzyl) called vittatin (51), for which a putative biosynthesis was suggested.

E.; Metzl, Nicolas; Olsen, Are; Sabine, Christopher L.; Heimann, M.;International audience; A temporally and spatially resolved estimate of the global surface-ocean CO2 partial pressure field and the sea-air CO2 flux is presented, obtained by fitting a simple data-driven diagnostic model of ocean mixed-layer biogeochemistry to surface-ocean CO2 partial pressure data from the SOCAT v1.5 database. Results include seasonal, interannual, and short-term (daily) variations. In most regions, estimated seasonality is well constrained from the data, and compares well to the widely used monthly climatology by Takahashi et al. (2009). Comparison to independent data tentatively supports the slightly higher seasonal variations in our estimates in some areas. We also fitted the diagnostic model to atmospheric CO2 data. The results of this are less robust, but in those areas where atmospheric signals are not strongly influenced by land flux variability, their seasonality is nevertheless consistent with the results based on surface-ocean data. From a comparison with an independent seasonal climatology of surface-ocean nutrient concentration, the diagnostic model is shown to capture relevant surface-ocean biogeochemical processes reasonably well. Estimated interannual variations will be presented and discussed in a companion paper.

Knowledge about HNS monitoring using radar remote sensing is not as extensive as for oil spills; however, any progress on this issue would likely advance the monitoring of both chemical and oil-related incidents. To address the need for HNS monitoring, an experiment was conducted in May 2015 over the Mediterranean Sea during which controlled releases of HNS were imaged by a multifrequency radar system. The aim of this experiment was to establish a procedure for collecting evidence of illegal maritime pollution by noxious liquid substances using airborne radar sensors. In this paper, we demonstrate the ability of radar imagery to detect and characterize chemicals at sea. A normalized polarization difference parameter is introduced to quantify both the impacts of released product on the ocean surface and the relative concentration of the substance within the spill. We show that radar imagery can provide knowledge of the involved HNS. In particular, one can distinguish a product that forms a film on the top of the sea surface from another that mixes with seawater, the information that is critical for efficient cleanup operations.

Our first objective was to test the effect of dietary carbohydrates themselves (without modification of dietary protein intake) on hepatic glucose gene expression (taking into account the paralogs). The second aim was to research if two isogenic trout lines had different responses to carbohydrate intake, showing one with a better use dietary carbohydrates. Thus, we used two isogenic lines of rainbow trout (named A32h and AB1h) fed with either a high carbohydrate diet or a low carbohydrate diet for 12 weeks. We analysed the zootechnical parameters, the plasma metabolites, the hepatic glucose metabolism at the molecular level and the hormonal-nutrient sensing pathway. Globally, dietary carbohydrate intake was associated with hyperglycaemia and down regulation of the energy sensor Ampk, but also with atypical regulation of glycolysis and gluconeogenesis in the liver. Indeed, the first steps of glycolysis and gluconeogenesis catalysed by the glucokinase and the phospenolpyruvate carboxykinase are regulated at the molecular level by dietary carbohydrates as expected (i.e. induction of the glycolytic gck and repression of the gluconeogenic pck); by contrast, and surprisingly, for two other key glycolytic enzymes (phosphofructokinase enzyme – pfkl and pyruvate kinase – pk) some of the paralogs (pfklb and pklr) are inhibited by carbohydrates whereas some of the genes coding gluconeogenic enzymes (the glucose-6-phosphatase enzyme g6pcb1b and g6pcb2a gene and the fructose1-6 biphosphatase paralog fbp1a) are induced. On the other hand, some differences for the zootechnical parameters and metabolic genes were also found between the two isogenic lines, confirming the existence of genetic polymorphisms for nutritional regulation of intermediary metabolism in rainbow trout. In conclusion, our study determines some new and unexpected molecular regulations of the glucose metabolism in rainbow trout which may partly lead to the poor utilization of dietary carbohydrates and it underlines the existence of differences in molecular regulation of glucose metabolism between two isogenic lines which provides arguments for future selection of rainbow trout. Summary: Using isogenic lines, this study determines some new, unexpected molecular regulation of the glucose metabolism in rainbow trout, which may partly lead to the poor utilization of dietary carbohydrates.

Models predict that some portion of this release will be offset by increased production of Arctic and boreal biomass; however, the lack of robust estimates of net carbon balance increases the risk of further overshooting international emissions targets. Precise empirical or model-based assessments of the critical factors driving carbon balance are unlikely in the near future, so to address this gap, we present estimates from 98 permafrost-region experts of the response of biomass, wild␣re, and hydrologic carbon ␣ux to climate change. Results suggest that contrary to model projections, total permafrost-region biomass could decrease due to water stress and disturbance, factors that are not adequately incorporated in current models. Assessments indicate that end-of-the-century organic carbon release from Arctic rivers and collapsing coastlines could increase by 75% while carbon loss via burning could increase four-fold. Experts identi␣ed water balance, shifts in vegetation community, and permafrost degradation as the key sources of uncertainty in predicting future system response. In combination with previous ␣ndings, results suggest the permafrost region will become a carbon source to the atmosphere by 2100 regardless of warming scenario but that 65%–85% of permafrost carbon release can still be avoided if human emissions are actively reduced.
